The negative can be used in such a phrase, but it would be better formulated in a positve form, by using "ever" instead of "never"
The phrase would then be:
"Have you ever been to turkey?" (sounds interrogative)
If not it should read:
"Have you never been to Turkey" (sounds incredulous or ironic)
